Heung-min Son then had a strike from a similar distance saved by keeper Jeroen Zoet at the other end.
But you always felt Tottenhams tactic of playing out from the back - regardless of whether their players were marked - in a bid to pull PSV out of position was going to end with them shooting themselves in the foot.
And so it came to pass on the half-hour mark when, after a several narrow escapes, their last man Alderweireld was mugged by Lozano, who set off towards goal.
Ole Toby mug did well to get back and tackle the PSV ace as he pulled the trigger.
But unfortunately for him and Spurs all he succeeding in doing was deflecting the ball looping over Lloris and into the net.

Tottenham looked the only likely winners after the break as Christian Eriksen, playing his first game for a month after recovering from an abdominal injury, had a shot well saved by Zoet.
Their great Dane then turned provider with a succulent cross for Kane to head home the winner in the 54th minute.
The England captain has now scored 11 goals in his first 13 matches in the Champions League.
Sub Erik Lamela went close to adding a third for Tottenham with a shot that hit the bar.
Lloris turbulent season then continued as he was sent off for bringing down Lozano outside of his area in the 79th minute.
Sub keeper Michel Vorm, on for the sacrificed Son, tipped Pereiros resulting free-kick around the post with his first touch.
But there was nothing the Dutch stopper could do to keep out De Jongs close range flick for the leveller two minutes from time.
It means both teams have just one point from their opening three matches - and with Spurs still having to travel to Barcelona it does not look like being enough to take them through.
